Continuous Production of Cadaverine Using PANI/Ba-Alginate Enzyme Reactor

Abstract
The production of cadaverine (1,5-diaminopentane) can be achieved through the decarboxylation of L-lysine catalyzed by lysine decarboxylase (LDC). This enzyme require pyridoxal 5¡¯-phospate (PLP) as a cofactor to increase conversion and yield. In this work, immobilization of LDC with polyaniline nanofiber (PANI) and Ba alginate (PANI/Ba-alginate) was developed to improve production rates and stability. We also developed a continuous cadaverine production process using PANI/Ba-alginate enzyme reactor. The optimal conditions for cardaverine production were 4% alginate concentration and 0.5 M BaCl2. Developed PANI/Ba-alginate LDC composite was very stable and its enzyme activity was maintained over 90% for 6 days continuous operation. The continuous enzyme reactor (CSTR) was operated for 144 h with a constant flow rate and achieved continuous cadaverine production. When the residence time was 33 h, 86% cadaverine conversion was achieved from 300 mM of lysine.
